In the present study we identified and characterized the novel sHsp Hsp21 of the human fungal pathogen Candida albicans. Using a reverse genetics approach we demonstrate the importance of Hsp21 for

Small but crucial: the novel small heat shock protein Hsp21 mediates stress adaptation and virulence in Candida albicans Small heat shock proteins (sHsps) have multiple cellular functions. However, the biological function of sHsps in pathogenic microorganisms is largely unknown. In contrast, C. albicansHsp21 shares 96%, 53%, 40%, and 38% identity with uncharacterized proteins from Candida dubliniensis, Candida tropicalis, Candida parapsilosis, and Candida orthopsilosis, respectively, and 39% identity with the uncharacterized sHsp Hsp18 from Pichia stipitis(Figure 1and). HSP21 - Small heat shock protein 21 - Candida albicans (strain SC5314 / ATCC MYA-2876) (Yeast) - HSP21 gene & protein UniProtKB - Q5AHH4 (HSP21_CANAL) HSP21 orthologues are present in the majority of pathogenic Candida species, the gene is not found in humans. These results indicate that Hsp21 represents an attractive alternative to Hsp90 for combinatorial immunotherapeutic-antifungal treatment strat-egies. Results Phylogenetic relatedness of C. albicans Hsp21 and human sHsps Hsp21 was found in several pathogenic Candida species but not in humans.

This study investigated the signature profiles of C. albicans planktonic cells and biofilms in response to caspofungin (CAS) by mass spectrometry-based shotgun proteomics. We found that C. albicans biofilms were twofold more resistant to CAS with reference to planktonic BibTeX @MISC{Albicans_smallbut, author = {Candida Albicans and François L. Using a reverse genetics approach we demonstrate the importance of Hsp21 for resistance of C. albicans to specific stresses, including thermal and oxidative stress. • Candida spp are the fourth most common cause of hospital-acquired systemic infections in the United States with crude mortality rates of up to 50%.

Heat shock protein required for pathogenicity. Mediates thermotolerance and adaptation to oxidative stress and ethanol-induced stress. Required for invasive growth and filament formation under various filament inducing conditions. Plays a role in the capacity of damaging human-derived endothelial and oral epithelial cells during infection.

5 Dec 2020 Until recent years, Candida spp. had fundamentally been linked to cancerous processes as it is an opportunist pathogen that takes advantage of  SUMMARY. Candida albicans is the most prevalent human fungal pathogen, that is component of the Small Heat Shock Protein Hsp21 Mediates Stress Ad-. In the fungal pathogen Candida albicans, Hsp90 enables drug resistance and Small but crucial: the novel small heat shock protein Hsp21 mediates stress.

Our results indicate that combinatorial therapy targeting Hsp21, Hsp21 potentiates antifungal drug tolerance in Candida albicans. PLoS One, 8(3). Small heat shock proteins (sHsps) have multiple cellular functions. However, the biological function of sHsps in pathogenic microorganisms is largely unknown.

This prompted us to investigate the effects of a broad range of different antifungal drugs on an Hsp21-null C. albicans mutant strain. Our results indicate that combinatorial therapy targeting Hsp21, Hsp21 potentiates antifungal drug tolerance in Candida albicans.
